Affinity Proteomics and LC-MS synergize Biomarker Discovery, Characterization and Validation

Blood is an ideal, minimally invasive sample for biomarker discovery, but its complexity might make untargeted, deep proteomics challenging. This work compares plasma proteomics approaches using both Olink® Explore/Explore HT and LC-MS on the same samples. We analysed small cohorts of biofluids (urine, plasma, CSF, and dried blood spots) across both platforms, performing pathway analysis to highlight the complementarity of each data set. In a cohort of 40 COVID-19 patients, Olink Explore HT enabled precise measurement of cytokine storm markers, which are challenging for LC-MS, demonstrating the added depth provided by combining both technologies.
