Better resolution for CE-SDS analysis of new biopharmaceutical modalities through fine-tuning the separation temperature

Abstract
Biopharma researchers facing a difficult time adopting CE-SDS for purity/heterogeneity analysis of new protein modalities can now rapidly accommodate these analytes by simple, precise temperature adjustments to achieve the desired resolution. Since the electromigration property of any protein – SDS complex is an activated process and each and every single molecule have their own activation energy requirement, one can adjust the separation temperature to alter the separation performance for different samples. In other words, the separation between any proteins of interest can be easily fine-tuned by precisely varying the separation temperature in the PA 800 Plus Pharmaceutical Analysis System to attain the required resolution. SDS-CGE analysis of a biotherapeutics revealed no correlation between the activation energy requirement of the different species and their molecular weights, further emphasizing the importance of separation temperature optimization to obtain high resolution between the solute molecules of interest.
