EAD easy as 1-2-3: Making characterization of complex biotherapeutics routine

Abstract
Electron-based dissociation mechanisms have shown great promise for advanced characterization of biomolecules. However, routine adoption has been limited due to poor fragmentation efficiency, need for tuning and lack of reproducibility. To unleash this promise, SCIEX has recently released the ZenoTOF 7600 system mass spectrometer that includes electron-activated dissociation (EAD). EAD is unique because the cell produces an intense, free electron beam that can be used to efficiently fragment several different compound classes. As a result, the benefits of free electron fragmentation can be realized within peptide mapping experiments on LC timescales. EAD data can be processed and annotated using Protein Metrics, Inc. ByosTM software thereby enabling advanced characterization of various protein biotherapeutics within an automated and routine workflow. In this webinar, we will describe the underlying technology of EAD within the ZenoTOF 7600. We will then share examples of EAD fragment ion spectra processed using ByosTM for the site specific determination of glycosylation, characterization of disulfide bonded peptides, and discrimination of amino acid isomer pairs like isoaspartic acid from aspartic acid and leucine from isoleucine. Finally, we will showcase an example of in depth characterization of an antibody drug conjugate digest.
