Abstract

Isolation of tumor infiltrating leukocytes (TILs) and optimized workflow for faster and improved flow analysis

Recent advances in immuno-oncology research has proven the critical role of immune cells in cancer therapy. Deep understanding of the interaction and phenotype of immune and tumor cells within the tumor microenvironment is key to developing future immune based cancer therapy strategies. However, the most significant challenge to working with solid tumors is the need to dissociate the samples prior to their use in downstream applications such as immunomagnetic enrichment, flow cytometry and single-cell sequencing. Furthermore tumor cells can cause background signals and introduce bias to the analysis of immune cells.

Learning Objectives:

•    Optimized workflows for isolation and flow cytometric phenotyping of mouse TILs
•    Best tools and solutions to increase the quality of the data, reduce experimental time and ease of scale up during TIL analysis using flow cytometry
•    Best methods for reducing biases during TIL analysis
