Abstract

Little and Large - Using the flexibility of the MoFlo Astrios from sorting bacteria, through the more classic blood and human cells through to plant protoplasts!

Our MoFlo Astrios serves or multidisciplinary Department of Biology through our open access Bioscience Technology Imaging and Cytometry Core Lab at the University of York. As a core instrument with internal users with very varied demands with the added variety coming in from external users, it is vital that our cell sorter can cope with as diverse a range of samples types as possible.

The presentation is not simply about the samples, but how we approach sorting varied sample types to give us the best success rates for each samples type with tips and tricks along the way.

We will present data demonstrating the variety of cell sorts that have been undertaken. This will look at bacterial samples that are being selected for their fluorescent protein expression through to the large plant protoplasts. These samples require the range of nozzle sizes and very different pressure tolerances.  

We also discuss our immunology driven data sets that are being used for downstream RNA and Mass Spectrometry analyses followed by the need for sterile sorting for human prostate research and samples that require multiwall plate sorting and the strategies used to get near 100% success rates for single cell dispositions.
