QC Considerations for Flow Cytometry and Imaging Particles

Abstract
We will begin by introducing myself and Spherotech. Moving onto definitions of terms, particles and flow cytometer anatomy will be reviewed, as well as their common manufactured and data acquisition ranges. QC considerations are focused on alignment, calibration, and cross-calibration. Once Alignment is defined, we will explore the tools and protocols for data acquisition for alignment analysis and how to set an SOP for those measurables. Defining and outlining how to Calibrate and instrument will follow and what tools and parameters are used for the acquisition and determination therein. We will also define MEF and touch on other similar units. Lastly, we will exercise one QC fundamental - specifically Calibration, to then apply MEF values to unknown populations so end users can make statistically significant determinations when performing novel research, namely Cross Calibration. Low end sensitivity and background measurements by Q and B values will be reviewed in closing, providing an avenue for additional, optional QC.

  • Identify the tools needed and perform alignment for daily instrument QC, as well as produce Levy Jennings Graphs for CV and MFI tracking.
  • Identify the tools needed and perform instrument calibration and attain Calc. MEF, Rsq, and Residual Values
  • Cross calibrate MEF values and assign them to unknown populations.
