A Workflow for Knock-in Genome Editing: Simplified

    Biography
      Dr. Matthew Poling earned his Ph.D. in Biomedical Sciences from UC San Diego and joined Thermo Fisher Scientific in 2015. He is currently the lead Product Manager for genome editing technologies and has led the development of the TrueTag Donor DNA system, as well as the TrueDesign Genome Editor online tool. Matt is also responsible for developing and expanding the Invitrogen CRISPR-Cas9 and TALEN technology solutions and works closely with Thermo Fisher's research and development scientists to create innovative genome editing tools and services.

    Genome engineering technologies have revolutionized genetic research and dramatically accelerated the pace of discovery. While knockout editing has garnered much of the attention, efforts are increasing to build new tools for creating knock-in cell models in both immortalized cell lines and primary cells, such as T-cells and iPSCs. This seminar will highlight recent advancements in gene editing tools, including application data, that demonstrate the power and ease of a new platform to design genome editing experiments and how innovative donor DNA strategies can be combined with next-generation CRISPR-Cas9 tools and TAL effector nucleases.
     
    • Be able to choose which nucleases, Cas9 or TAL effector, are optimal for a particular knock-in editing experiments.
    • Recognize the caveats of different knock-in DNA donors sources 
    • Gain working knowledge of the TrueDesign™ Genome Editor tool to craft experiments for base insertions, deletions, SNP changes, and gene tagging
    • Discover how to achieve enrichment of up to 100% edited cells without cloning steps for gene knock-in experiments.
